comment on [Question] eGFR dropped 8 points during rapid loss, should i worry in c/bloodwork · -36 points · 4 days ago
Yes. Repeat before you react. Almost every alarming single value I have posted here regressed on the repeat.
comment on [Concerning] ferritin 9. i was eating 1,100 calories and calling it discipline. in c/bloodwork · 4 points · 5 days ago
Yes — same lab, same fasting state, same rough time of day, or the comparison is doing nothing.
comment on [Results] hs-CRP halved before the scale moved much in c/bloodwork · 91 points · 11 days ago
Added ApoB to the panel after a thread here. It told me something the standard lipid panel had been hiding.
comment on [Discussion] glucagon is doing more work than we give it credit for in c/retatrutide · 15 points · 24 days ago
What the glucagon arm is doing, as best anyone can say from public data.
Disagree with this specific inference. A 48-week readout does not tell you the shape of the curve after it.
comment on my resting HR went from 58 to 66 on titration and settled back in c/tirzepatide · 2 points · 24 days ago
2.5mg is a four-week starting dose, not a maintenance dose.
Not sure about this bit. The GIP arm being real does not tell you that it is what caused your particular week.
comment on [Question] does the vial concentration change how much dead space costs me in c/tirzepatide · 90 points · 27 days ago
Vials and pens carry the same molecule; what differs is fill volume, device tolerance and whether you are doing your own arithmetic. Neither is inherently more accurate.
comment on week 67 check-in — 10kg down, muscle cramps manageable, one thing confusing me in c/bloodwork · 0 points · 27 days ago
This. Reference ranges are population intervals, not targets, and conflating them causes a lot of unnecessary alarm here.
comment on anyone else notice retatrutide kicking in around week 83 in c/retatrutide · 71 points · 29 days ago
Small fix — three receptors, not two. GLP-1, GIP and glucagon, and the third one is the reason this compound gets its own board.
comment on [Discussion] the ALT advice in here is 2 years out of date in c/bloodwork · 27 points · 1 months ago
same lab, same conditions, or you are comparing noise
Agreed — and standardising the draw conditions is the cheapest improvement available.
comment on [Discussion] the ALT advice in here is 2 years out of date in c/bloodwork · 48 points · 1 months ago
Yes — same lab, same fasting state, same rough time of day, or the comparison is doing nothing.
comment on retatrutide is the most under-discussed thing on this board in c/retatrutide · 27 points · 1 months ago
the glucagon component is why the metabolic story reads differently
comment on [Discussion] we are measuring tirzepatide at the wrong time and calling it noise in c/tirzepatide · 7 points · 1 months ago
Held 7.5 for five months. Everyone kept asking when I was going to 10. The answer was never, because 7.5 was working.
comment on retatrutide is the most under-discussed thing on this board in c/retatrutide · 64 points · 1 months ago
The thing that surprised me was how much of the discussion here is inference rather than measurement.
comment on small win: glucagon stopped being a problem at week 32 in c/retatrutide · 1 points · 1 months ago
Push back: quoting the phase 2 headline as an expected outcome is not a fair reading of a small trial with wide intervals.
comment on how much of what we believe about Mounjaro actually comes from Zepbound threads in c/tirzepatide · 4 points · 2 months ago
Agreed, and the trial number is a mean of a very wide distribution — the tails are enormous and nobody posts from the middle.
comment on [Discussion] we are measuring Zepbound at the wrong time and calling it noise in c/tirzepatide · 181 points · 2 months ago
Weekly dosing again, half-life in the same neighbourhood as sema, so a step change takes about a month to reach steady state. Judging a new step at day five is judging noise.
comment on Zepbound — my 8-week log, condensed into one table in c/tirzepatide · 40 points · 3 months ago
Yes. I stepped to 12.5 because the calendar said so, not because anything needed fixing, and I regretted it for a fortnight.
comment on unpopular opinion: most of what gets said here about bloodwork is guesswork in c/bloodwork · 554 points · 4 months ago
What did the repeat show?
comment on Mounjaro: the version I wish someone had shown me in week 1 in c/tirzepatide · 2 points · 4 months ago
The 21% figure is a 72-week mean on the highest arm. Quoting it as what someone should expect by week 39 is not a fair reading.
comment on is it normal to get fatigue at week 5 in c/retatrutide · 130 points · 6 months ago
comparing reta phase 2 to sema phase 3 is comparing different things
Agreed, and the glucagon arm is exactly why the comparison threads do not work.
comment on help me understand glucagon, I have read the wiki twice in c/retatrutide · -23 points · 7 months ago
The phase 2 readout ran to 48 weeks in a few hundred participants with a slow escalation. Both the size and the duration matter when people quote the headline number.
comment on [Discussion] ApoB is doing more work than we give it credit for in c/bloodwork · 13 points · 8 months ago
Different laboratories use different assay platforms with different calibration. Comparing across labs adds a systematic offset that is invisible on the report.
comment on 14 months in and retatrutide is still the thing I get wrong in c/retatrutide · 1 points · 8 months ago
the phase 2 numbers were striking and they were also 48 weeks in a small population
honest_syringe33 is right about the escalation being the variable. It explains most of the difficult reports here.
comment on 14 months in and retatrutide is still the thing I get wrong in c/retatrutide · 219 points · 8 months ago
small trial, big effect, wide error bars
comment on my A1c moved and I cannot work out whether TRIUMPH is why in c/retatrutide · 57 points · 9 months ago
Increases in heart rate have been reported across this receptor class. The magnitude and clinical significance are exactly what phase 3 is powered to establish.
comment on someone explain 15mg to me like I have not read a paper in years in c/tirzepatide · 76 points · 1 year ago
Push back: "gentler than sema" is a population statement. Plenty of people on this board have the opposite experience and they are not doing it wrong.
comment on [Results] 4 weeks on 15mg, full numbers, ask me anything boring in c/retatrutide · 524 points · 1 year ago
This. The phase 2 result was genuinely large and it was also 48 weeks in a few hundred people.
comment on how much of what we believe about Mounjaro actually comes from SURMOUNT threads in c/tirzepatide · 26 points · 1 year ago
I would separate the two claims. That the GIP arm exists is uncontroversial; that it explains your fatigue pattern is a guess.
comment on 5 months in and dose escalation is still the thing I get wrong in c/retatrutide · 19 points · 1 year ago
Correction: TRIUMPH is the phase 3 programme.
This is the sentence the rest of the board should read first. Phase 2 is not a label.
comment on [Meta] proposal — a flair for SURMOUNT posts in c/tirzepatide · 875 points · 1 year ago
The distribution matters more than the mean. In the trial population the interquartile spread was wide enough that two honest people can have completely different runs on the same arm.
comment on [Lab] 9th independent test on QYB — 97.7% on a claimed 97.0%, and the trend is the interesting part in c/retatrutide · 1 points · 1 year ago
Glucagon receptor agonism is associated with increased energy expenditure and with hepatic effects. That is a mechanistic story with strong preclinical support and limited phase 2 human data.

Substantial weight loss independently moves lipids, liver enzymes and several other markers. Attributing a change to a compound while losing weight is confounded by design.

Pen or vial? The step sizes available differ and it changes this answer.

not approved anywhere, which is the single most important fact in this board
comment on 3 months in and triple agonist is still the thing I get wrong in c/retatrutide · 211 points · 1 year ago
Are you comparing against phase 3 numbers for something else? They are not comparable.
