a rough fortnight at each step is normal, a rough three months is not

Correction: that rule applies to the vendor boards specifically, not site-wide. The wording is on the rules page.

Same lane, four orders, 14 to 16 days, all four within a couple of days of the quote. That consistency is the actual finding.

Correction: the starting dose is a tolerance step rather than a treatment dose. The distinction is the answer to the question above.

Titration is a tolerance ladder. Each step exists so the gastrointestinal effects settle before the next increase, which is why the intervals are what they are.

Agreed. Publishing the ranking formula is the difference between a feed and a black box, and it costs nothing to do.

comment on 13 months in and basics are still the thing I get wrong in c/newbiequestions · 336 points · 1 year ago

Research-use-only material is not approved for human use. That is a plain statement of regulatory fact and it belongs in every beginner thread.

phase 2 finds a dose, phase 3 measures the effect

Push back: comparing a July transit with a February one on the same lane is not a comparison.

nobody warns you about how much of this is timing rather than dose

A certificate of analysis is a document describing one batch: what was tested, by what method, and what the result was. Its value depends entirely on whether it names the batch and the method.

Yes. Holding the dose is a real option and it does not mean you have failed at anything.

Are you comparing against a GLP-1 monotherapy result? They are not comparable.

holding a dose an extra month is a legitimate answer to bad tolerance

I would not compare your numbers to that population. Different baseline, different regimen, different trial.

Same. The injection was the thing I was most nervous about and the thing that turned out to matter least.

comment on help me understand SURMOUNT, I have read the wiki twice in c/trialwatch · 74 points · 1 year ago

Started keeping the trial identifiers straight in a note file because I kept mixing up two programmes in the same sentence.

I would not read a single slow shipment as a lane change. Post it and let the median move if it is real.

Careful with the supplement recommendation. There is no evidence for it here and the mechanism you are proposing does not hold.

Correction: SURMOUNT is the obesity programme and SURPASS is the diabetes one. The figure you quoted belongs to the other one.

Intention-to-treat analyses everybody randomised regardless of what they did afterwards. Completer analyses only those who finished. The second is systematically more flattering and both are legitimate if labelled.

Yes — the interval is the finding. A point estimate with a wide interval is a hypothesis in a nice font.

comment on my TSH moved and I cannot work out whether sulphur burps are why in c/sideeffects · 23 points · 2 years ago

Yes.

Agreed, with the caveat that "usually settles" is not "always settles" and the exceptions should still get asked about properly.

The ranking functions are published: the hot ordering combines score magnitude with age, and the controversial ordering weights total volume by the balance between up and down votes. Both are reproducible from the numbers on each submission.

Split one order across two months on purpose to compare summer and winter. Four days apart, same carrier, same route.

permalink

Correction: that is a known effect of reduced intake rather than a direct drug effect. The distinction matters because the fix is different.
