Overview — page 3 of 3

comment on what would you tell week-1 you about non-peptide in c/orforglipron · 54 points · 12 months ago

Oral semaglutide is a peptide co-formulated with an absorption enhancer and carries food and water timing requirements. A small molecule does not have that constraint, which is the practical distinction.

Spent an evening on the histology scoring system and understood the trial literature far better afterwards.

Correcting myself: the readout I quoted was the 55-week interim rather than the endpoint.

Agreed on keeping every form and date.

Agreed — write the application against the published criteria rather than around them.

Registration by the therapeutic goods regulator establishes that a product may be supplied. Subsidy listing is a separate decision about public funding, with its own criteria and its own timetable.

comment on [Meta] proposal — a flair for SURMOUNT posts in c/tirzepatide · 1 points · 1 year ago

Right. And the sema-to-tirz "conversion" people post is invented. There is no published equivalence.

satiety signalling rather than incretin signalling

permalink

comment on why does nobody talk about cagrilintide in c/cagrilintide · 172 points · 1 year ago

Amylin is co-secreted with insulin and acts on satiety and gastric emptying through its own receptor complexes.

Adding the standing caveat — nothing here is approved standalone and research material is not for human use.

comment on [PSA] gallbladder is not what most of this community thinks it is in c/sideeffects · 16 points · 1 year ago

Kept a one-line-a-day log for two months.

This is the shape almost everybody here describes, and it is the single most reassuring thing in the thread.

Is it every week or did it start at a particular dose?

Correction: that is a known effect of reduced intake rather than a direct drug effect. The distinction matters because the fix is different.

comment on [Discussion] we are measuring Mounjaro at the wrong time and calling it noise in c/tirzepatide · 373 points · 1 year ago

SURMOUNT-1 ran 72 weeks with the top arm around 21% mean body weight change. SURPASS is the diabetes programme and reports glycaemic endpoints, so quoting the two interchangeably is a category error.

if it is severe or persistent that is a clinician question, not a board question

Not convinced. A warm pack on day six is expected behaviour from a pack designed for four.

Careful with the supplement recommendation. There is no evidence for it here and the mechanism you are proposing does not hold.

Yes — a logger in the box turns a fortnight of speculation into a chart.

Agree. The GIP component is the interesting half and it barely gets discussed outside c/glp1science.

Hair shedding after significant weight loss is a telogen effect that follows the rate of loss. It is not specific to these compounds and it resolves.

comment on [Question] special authorization — what am I missing here in c/glp1canada · 62 points · 1 year ago

Quantity limits restrict how much may be dispensed in a period even where coverage exists.

karim_restrepo is right that a rejected claim and a denied authorization are different animals.

That is the ambient temperature at the destination, not the temperature inside the box. The logger measures the second one.
