comment on [Discussion] are the c/vendorvetting evidence rules too strict (no) in c/meta · 1 points · 5 days ago

The evidence standard on the vendor boards requires a document, a date or a named service behind any claim about a supplier. It is what makes those threads worth reading years later.

regain after stopping is the expected outcome, not a personal failure

comment on [Milestone] first time in a decade my blood pressure is normal in c/progresspics · 6 points · 15 days ago

This. Waist measurement kept moving through my longest stall and it was the only thing that kept me sane.

permalink

comment on [Discussion] we have no vocabulary for success that is not a number going down in c/maintenancephase · -8 points · 18 days ago

Stretched from seven to ten days and then to twelve over about four months. Appetite told me where the limit was before the scale did.

comment on [Results] two years, weight stable within 3kg, dose at a quarter of peak in c/maintenancephase · 5 points · 20 days ago

Adding one thing — decide what maintenance looks like before you arrive at it.

This reframing is the most useful thing on the board for anyone approaching this phase.

Stalled for 23 weeks and measured my waist out of stubbornness.

india_generic_g is right that the clothes tell you before the scale does.

Small correction, kindly meant: that is a range rather than a regain. Weight moves day to day in everybody.

How long was the stall before it moved?

comment on [Results] 79 weeks, 21kg, and non-peptide was the hard part in c/orforglipron · 43 points · 24 days ago

Oral semaglutide is a peptide co-formulated with an absorption enhancer and carries food and water timing requirements.

piotr_bruun is right that milligram comparisons across classes tell you nothing.

phase 3 is where the comparison becomes fair

oral semaglutide is a peptide with an absorption enhancer, this is not that

comment on why does nobody talk about non-peptide in c/orforglipron · 13 points · 2 months ago

This. The absence of food and water timing restrictions is the practical difference people will actually notice.

permalink

comment on why does nobody talk about non-peptide in c/orforglipron · 18 points · 2 months ago

Went looking for independent testing on this and found essentially nothing, which was clarifying.

comment on week 95 check-in — 48kg down, nausea manageable, one thing confusing me in c/maintenancephase · 36 points · 4 months ago

the habits are the part that has to outlive the dose

comment on genuine question about oral GLP-1 that I am slightly embarrassed to ask in c/orforglipron · 40 points · 4 months ago

do not assume reconstitution intuitions apply, there is nothing to reconstitute

permalink

comment on genuine question about oral GLP-1 that I am slightly embarrassed to ask in c/orforglipron · 52 points · 4 months ago

Spent an evening reading about small-molecule agonism at a peptide receptor. Genuinely interesting engineering.

Phase 2 or phase 3?

permalink

comment on [Question] struggle — what am I missing here in c/progresspics · 109 points · 4 months ago

Circumference measurements often continue changing during a scale plateau because composition changes are not visible on a bathroom scale.

ask what the reference standard was

permalink

comment on [Discussion] lowest effective dose is doing more work than we give it credit for in c/maintenancephase · 19 points · 6 months ago

Agreed — and appetite is the leading indicator.

Disagreeing with this bit: a two-kilo range is not drift and treating it as a problem creates one.

Storage guidance that names a temperature, a state and a duration reflects actual stability work. Generic guidance usually reflects a template.

Correction: that trial measured regain after withdrawal, not failure of the drug during treatment. Very different claim.

permalink

comment on small win: non-peptide stopped being a problem at week 82 in c/orforglipron · 101 points · 8 months ago

Because it is not a peptide, it is not subject to the same degradation pathways, does not require the absorption enhancers used for oral peptides, and can be formulated as a conventional tablet.

permalink

comment on [Question] how do you actually verify research peptides in c/researchpeptides · 25 points · 9 months ago

That method was developed for a different compound class and would not resolve the impurities you are asking about.

My goal weight at week one was a number I made up. I have revised it twice since and I am happier for it.

comment on is it normal to get early fullness at week 90 in c/maintenancephase · 322 points · 12 months ago

Careful — a dose reduction plan is a conversation with someone who knows your history, not a board consensus.

permalink

comment on what changed for me between month 3 and month 6 in c/orforglipron · 132 points · 1 year ago

Milligram comparisons across molecule classes are meaningless. Potency is a property of the molecule at its receptor, not of the number on the label.
