comment on [Explainer] area% is not mass% and here is exactly why it matters in c/coa · 12 points · 6 days ago

Asked for the storage condition behind the stability statement. Got it in a sentence. It is a two-line email and almost nobody sends it.

comment on [Suspicious] identical chromatogram on two different compounds from the same vendor in c/coa · 46 points · 8 days ago

Water content 3.8%, acetate 6.2%, purity 99%, net peptide about 90% of label weight. All four numbers honest, and I had to read it twice.

comment on [COA] what does "related substances 1.4%" actually mean for my vial in c/coa · 39 points · 11 days ago

Asked TFC for the batch-specific certificate and had the full document — batch, date, method line naming the column — inside six hours. That is now my baseline for everybody.

Has a peer-to-peer been offered or requested?

vikram_mbeki is right that this is documentation rather than persuasion. It took me a year to accept that.

Careful — "the metadata looks odd" is a reason to ask a question, not a conclusion. Re-issues are ordinary.

net peptide content and purity are two different numbers on the same vial

comment on genuine question about area percent that I am slightly embarrassed to ask in c/coa · 63 points · 2 months ago

Pushing back a little: a missing field is usually presentation, not concealment. Ask for the full document before drawing conclusions.

comment on help me understand COA, I have read the wiki twice in c/coa · 138 points · 2 months ago

That is not what the method line says.

india_generic_g has the distinction right — one is a fraction of the detector signal, the other is a fraction of the mass.

Disagree that a lower number is automatically worse. A conservative claim that survives testing is worth more than an optimistic one that does not.

Internal appeals are decided by the plan. External review is decided by an independent body and, where it applies, its determination is binding. They are separate mechanisms and the second is chronically underused.

comment on [Question] certificate of analysis — what am I missing here in c/coa · 3 points · 3 months ago

That is not what the method line says. It names the technique; it does not certify the operator.

comment on unpopular opinion: most of what gets said here about external review is guesswork in c/insurancefights · 93 points · 3 months ago

Careful — the deadline runs from the date on the letter. Waiting for a call back can cost you the appeal entirely.

Asked for the storage condition behind the stability statement.

Agreed, and the practical version is: ask for the itemised table. If it exists, you get it.

ask for the storage condition the stability statement was written against

Employer-sponsored plans may be regulated differently from individually purchased ones, which changes both the appeal route and which regulator hears

Agreed — and request the bulletin by number. They have to give it to you.

Peer-to-peer took fifteen minutes and resolved something a written appeal had been sitting on for a month.

Related substances itemised individually is a much stronger document than a single total, because it tells you the shape of the impurity profile rather than its size.

net peptide content and purity are two different numbers on the same vial

permalink

Same view. SWB prints the column and the gradient, which means somebody could actually reproduce the run.

That is not what the method line says. It names the technique; it does not certify the operator.

Correction: that is net peptide content, not purity. Different number, different meaning, and the document says so if you read the row label.

That advice is jurisdiction-specific and this board spans several. Say where you are.

HPLC-UV and LC-MS answer different questions

permalink

comment on [Discussion] the purity advice in here is 3 years out of date in c/coa · 38 points · 1 year ago

LC-MS gives you identity; UV purity gives you a relative quantity. A document with the first and not the second, or the reverse, is answering half the question.

Yes. The structure of the document matters at least as much as the number printed on it.

comment on [Lab] split one vial across Janoshik and Medutest — 98.1% and 97.6% in c/coa · 1 points · 1 year ago

Small fix — 214nm, not 280nm. It matters here because the impurity you are asking about barely absorbs at the longer wavelength.

permalink

comment on [Question] how do you actually verify formulary in c/insurancefights · 0 points · 2 years ago

Small fix — external review is independent of the plan. The second-level internal appeal is not.

permalink

comment on purity — 3 things I got wrong before I got it right in c/coa · 108 points · 2 years ago

Cosigning. The batch identifier is load-bearing, and a certificate that cannot be tied to the vial in your hand is not about your vial.
