Read the primary paper after arguing about the summary for a fortnight. The discussion section said almost the opposite of what the thread had concluded.

That study was in a rodent model. Worth stating, since the thread has been reading it as human data.

comment on why do people talk about units and mg like they are different things in c/newbiequestions · 96 points · 29 days ago

Appetite change and weight change are different measurements on different timescales. The first is usually noticed first and the second lags it.

Agreed.

This is the answer. Everything else in the thread is elaboration on it.

Yes — four nations, four pathways, and threads that ignore that give people the wrong answer confidently.

Reduced appetite makes hitting a protein target a logistics problem rather than a motivation problem, which is why the practical advice here is about

Adding the free proxy — track a few lifts monthly. It costs nothing and it is longitudinal.

The injection was scarier in my head than in reality. It took about ten seconds and I felt slightly silly afterwards.

comment on [Discussion] can we stop arguing about regain until somebody posts a number in c/tapering · 17 points · 2 months ago

Withdrawal studies across this class consistently show substantial regain after discontinuation, with appetite typically returning before weight changes. That is a description of the pharmacology of a chronic condition.

Gently disagree with the framing — nothing has gone wrong. Week 2 looking flat is the ordinary experience.

That shortage notice was national and does not apply where the question is coming from.

comment on someone explain appetite return to me like I have not read a paper in years in c/tapering · 19 points · 5 months ago

Edited a comment framing regain as a moral failure.

Disagreeing with this bit: stretching and reducing are different things and the post treats them as one.

Small fix — you have described interval stretching and called it a taper. The exposure profiles differ.

Not convinced by the linearity assumption. Dose-response in this class is not linear and the trials show it.

comment on [Question] the UK — has anyone got a straight answer on reimbursement in c/glp1eu · 20 points · 7 months ago

Cross-border prescription recognition exists under EU rules but is conditional and narrower than commonly assumed, with requirements about the prescription’s form and content.

tolerance to the gastric effect develops, appetite effect largely persists

Correcting my own comment: I attributed that to the GIP arm and the paper attributes it to the GLP-1 arm.

Cosigning on GIP. The genuinely interesting thing is that the biology is not settled and the clinical result is nonetheless robust.

Same view. Tolerance developing to the gastric effect while the appetite effect persists explains most of what the side-effect board reports.

This. Albumin binding and modification are why the half-life is what it is, and it is a design decision rather than an accident.
