comment on week 73 check-in — 28kg down, early fullness manageable, one thing confusing me in c/sideeffects · 32 points · 2 months ago

holding a dose an extra month is a legitimate answer to bad tolerance

This. Intention-to-treat versus completer analysis routinely moves the headline by several points.

How long was the randomised phase before any extension?

comment on [PSA] cold chain is not what most of this community thinks it is in c/coldchain · 36 points · 2 months ago

Correction: that stability figure covers the lyophilised powder, not the reconstituted solution. Different claim entirely.

comment on dose escalation is the most under-discussed thing on this board in c/retatrutide · 4 points · 2 months ago

Glucagon receptor agonism is associated with increased energy expenditure and with hepatic effects. That is a mechanistic story with strong preclinical support and limited phase 2 human data.

Open-label extensions lose their randomisation. Anybody still enrolled at week 104 is a selected group and the numbers describe that group.

comment on [Meta] the excursion rule is doing its job and people should stop complaining in c/coldchain · -12 points · 2 months ago

Ordered from the warehouse in Denmark instead of origin and the transit went from a fortnight to 7 days. Entirely different proposition in August.

comment on [Lab] TFC cagri — Janoshik came back 98.1% against a claimed 98.0% in c/cagrilintide · 44 points · 3 months ago

That result is preclinical. Worth flagging, since the thread has been reading it as human data.

Combination and monotherapy arms must be read separately.

nl_verzekering is right that the evidence base here is thin. Conclusions should be held loosely.

comment on 8 months in and amylin is still the thing I get wrong in c/cagrilintide · 0 points · 3 months ago

Nothing containing this compound is approved as a standalone product, and research-use-only material is not approved for human use.

comment on [Meta] proposal — a flair for co-agonism posts in c/cagrilintide · 14 points · 3 months ago

Small fix — amylin analogue, not a GLP-1 analogue. The whole mechanism argument changes on that word.

Kept fingersticks alongside the sensor for two weeks to sanity-check it. Worth doing once.

comment on genuine question about TB-500 that I am slightly embarrassed to ask in c/researchpeptides · 2 points · 3 months ago

Careful — that is a claim about efficacy and there is essentially no human evidence behind it.

a rough fortnight at each step is normal, a rough three months is not

sulphur burps are unpleasant and they are not dangerous

What dose and how many weeks at it before this started?

permalink

comment on [Results] 19 weeks, 7kg, and food noise was the hard part in c/semaglutide · 2 points · 4 months ago

My stall was eleven weeks. Eleven. Then it moved again without me changing anything at all.

Minor fix — the half-life is about a week, not about a day. The rest of your point stands.

Nothing on this board is legal advice. Where a notice carries a deadline or a consequence, that is a matter for somebody qualified in your own jurisdiction.

hub_ops is right about the programme names.

Disagreeing with this line: that is a relative reduction and the absolute numbers are considerably less dramatic.

Push back: quoting the phase 2 headline as an expected outcome is not a fair reading of a small trial with wide intervals.

if it is severe or persistent that is a clinician question, not a board question

permalink

comment on what would you tell week-1 you about reflux in c/sideeffects · 26 points · 7 months ago

A symptom that begins the day after the shot and fades by day four tracks the exposure curve. One that appears at random in week 65 probably does not, and that is worth saying to a clinician rather than to us.

Push back: handling advice from one compound does not transfer to another just because both are peptides.

Yes. Week 37 is when almost everybody writes the "is it still working" post, and almost everybody is fine.
