comment on [Paper] survodutide MASH data is the most interesting hepatic readout in the class in c/survodutide · 4 points · 1 days ago

This. Biopsy-confirmed and imaging-suggested are different evidentiary categories and get run together constantly.

What is intake actually looking like — fibre and water especially?

permalink

comment on what is bac water and why does everyone keep saying it in c/newbiequestions · 28 points · 5 days ago

Agreed — week two with nothing on the scale is so common it should be in the sidebar. It probably is.

Keeping a log, and what to put in it.

Agreed, with the caveat that "usually settles" is not "always settles" and the exceptions should still get asked about properly.

the first fortnight after a step is the worst of it, then it settles

comment on [Question] does the glucagon arm explain the heart rate signal in c/survodutide · 0 points · 7 days ago

Histological endpoints in this field are scored on biopsy: resolution of steatohepatitis without worsening of fibrosis, or improvement in fibrosis without worsening of steatohepatitis. Both are harder to achieve and to interpret than an imaging surrogate.

comment on [Seizure] third one in two years. the pattern is the carrier, not the country. in c/customs · 6 points · 9 days ago

ask the supplier what documentation accompanies the shipment

permalink

comment on [Discussion] every answer here is jurisdiction-specific and we keep forgetting in c/customs · 2 points · 13 days ago

the answer for New Zealand does not apply anywhere else

permalink

comment on [Failure] vendor vanished mid-order with 11 people waiting in c/darkspot · 45 points · 17 days ago

Push back: you did not lose that money by being stupid. You lost it to a shop with nothing verifiable on file, which is a different thing.

permalink

comment on [Regret] told my whole family, then stopped, then had to tell them that too in c/darkspot · 24 points · 21 days ago

Are you being harder on yourself than you would be on somebody else posting this?

permalink

comment on everyone here has a spreadsheet, do i need a spreadsheet in c/newbiequestions · 21 points · 22 days ago

Spent my first fortnight comparing myself to the loudest posters. Do not do that; they are not the middle of the distribution.

permalink

comment on the gallbladder question that gets asked weekly, answered properly in c/sideeffects · 22 points · 1 months ago

fatigue plus low intake is usually just low intake

permalink

comment on the hypoglycaemia question that gets asked weekly, answered properly in c/t2dglp1 · -14 points · 1 months ago

Incretin-based agents stimulate insulin secretion in a glucose-dependent manner, which is why hypoglycaemia risk with them alone is low. Risk rises with agents that act independently of glycaemia.

comment on [Lab] GGPeps survo — VendorInvestigate came back 97.9% against a claimed 97.0% in c/survodutide · 2 points · 1 months ago

Small fix — dual agonist, GLP-1 and glucagon. No GIP arm in this molecule.

permalink

comment on [Lab] GGPeps survo — VendorInvestigate came back 97.9% against a claimed 97.0% in c/survodutide · 3 points · 1 months ago

Which endpoint — histological response, fibrosis improvement, or fat fraction?

permalink

comment on [Lab] GGPeps survo — VendorInvestigate came back 97.9% against a claimed 97.0% in c/survodutide · 9 points · 1 months ago

Is that a weight number from a different programme?

permalink

comment on three years of glucagon threads, summarised so you do not have to read them in c/survodutide · 10 points · 1 months ago

Yes — histological endpoints are a far harder bar than the surrogate measures people are used to quoting.

permalink

comment on [Meta] the hypoglycaemia rule is doing its job and people should stop complaining in c/t2dglp1 · 9 points · 2 months ago

the fasting number is the one people fixate on and it moves last

permalink

comment on why does nobody talk about dual agonist in c/survodutide · 29 points · 2 months ago

read the histology endpoint definitions before quoting a response rate

permalink

comment on unpopular opinion: most of what gets said here about what went wrong is guesswork in c/darkspot · 6 points · 3 months ago

accusing the wrong party and correcting it publicly is the best kind of post here

permalink

comment on stalled for 14 weeks at 12.5mg and I refuse to panic this time in c/darkspot · 49 points · 3 months ago

somebody will read this at the point you were at, and it will help

permalink

comment on anyone else notice failure kicking in around week 21 in c/darkspot · 111 points · 3 months ago

Not convinced that was avoidable. Some of this is genuinely bad luck and treating everything as a lesson is its own trap.

permalink

comment on [Question] how do you actually verify gallbladder in c/sideeffects · 223 points · 3 months ago

How long after a step up did this start?

permalink

comment on [Meta] proposal — a flair for lowest effective dose posts in c/maintenancephase · 4 points · 4 months ago

Appetite returning before the scale moves follows from the mechanism: the appetite effect is the proximal one and weight is the lagging integrated outcome.

permalink

comment on anyone else notice fatigue kicking in around week 46 in c/sideeffects · 73 points · 4 months ago

a rough fortnight at each step is normal, a rough three months is not

permalink

comment on unpopular opinion: most of what gets said here about import is guesswork in c/customs · 27 points · 4 months ago

Where a supplier holds regional stock, the shipment is domestic and there is no import event at all. That is the structural fix rather than a workaround.

permalink

comment on the dose stretching thing finally clicked for me and I want to write it down in c/maintenancephase · 126 points · 4 months ago

Wrote down what maintenance would look like at about week 86, before I needed it. Having decided in advance made the transition boring.

permalink

comment on the dose stretching thing finally clicked for me and I want to write it down in c/maintenancephase · 273 points · 5 months ago

None of this is a taper plan. What dose holds for you is a question for somebody with your history in front of them.

permalink

comment on [Question] how do you actually verify glycaemic control in c/t2dglp1 · 1 points · 5 months ago

That A1c change is inside the assay’s variability and the interval you measured over is too short.

permalink

comment on why does nobody talk about lost money in c/darkspot · 26 points · 5 months ago

Chased the scale instead of the measurements for a year and made myself miserable for no additional progress.

permalink

comment on my ApoB moved and I cannot work out whether beginner is why in c/newbiequestions · 17 points · 5 months ago

That is area percent from a certificate, not the amount of peptide in the vial. Two different numbers and a very common mix-up.

permalink

comment on three years of survodutide threads, summarised so you do not have to read them in c/survodutide · 38 points · 5 months ago

Push back: the evidence here is phase 2, in a specific population, and it does not support a general ranking.

permalink

comment on the jurisdiction thing finally clicked for me and I want to write it down in c/customs · 11 points · 5 months ago

Not convinced. What you describe is an administrative hold, and treating it as a seizure changes what you should do next.

permalink

comment on stalled for 23 weeks at 10mg and I refuse to panic this time in c/newbiequestions · 12 points · 5 months ago

Yes. The starting dose is a tolerance step. It is not supposed to do very much.

permalink

comment on MASH — my 24-week log, condensed into one table in c/survodutide · 2 points · 5 months ago

MASH endpoints are histological, which is a much harder bar

permalink

comment on Germany: personal import, and what it actually costs here in c/customs · 1 points · 6 months ago

Correction: that is a request for information, not a seizure notice. Different document, different process, different deadline.

permalink

comment on [Meta] the gallbladder rule is doing its job and people should stop complaining in c/sideeffects · 252 points · 6 months ago

On buying things to fix side effects, which I did extensively and would not do again.

This is the shape almost everybody here describes, and it is the single most reassuring thing in the thread.

permalink

comment on tried week one for 18 weeks. here is what happened. in c/newbiequestions · 8 points · 6 months ago

the starting dose is not meant to be the dose that works

permalink

comment on small win: goal weight stopped being a problem at week 19 in c/maintenancephase · 23 points · 6 months ago

maintenance is a phase, not a finish line
