comment on the GIP question that gets asked weekly, answered properly in c/tirzepatide · 32 points · 11 months ago

The 21% figure is a 72-week mean on the highest arm. Quoting it as what someone should expect by week 34 is not a fair reading.

Lyophilisation removes the water that drives most degradation pathways. That is why the dry cake is comparatively robust and the reconstituted solution is not.

Weekly dosing again, half-life in the same neighbourhood as sema, so a step change takes about a month to reach steady state. Judging a new step at day five is judging noise.

Some chains require a memo or destination tag to route funds to the right account. Omitting it on those chains is one of the more common ways payments go astray.

That is a waist figure in the weight column. Easy mistake and it makes the chart look dramatic.

The sulphur burps profile felt different rather than worse. Hard to describe and impossible to quantify, so take it as an impression.

comment on stalled for 8 weeks at 15mg and I refuse to panic this time in c/dosinglogs · 9 points · 1 year ago

Yes — log what happened rather than what you intended. My retrospective entries were quietly wrong every time.

comment on triple agonist: the version I wish someone had shown me in week 1 in c/retatrutide · 1 points · 1 year ago

Push back: quoting the phase 2 headline as an expected outcome is not a fair reading of a small trial with wide intervals.

Not convinced.

This is the rule for anything unusual. Identity before anything else.

comment on unpopular opinion: most of what gets said here about pharmacology is guesswork in c/glp1science · 23 points · 1 year ago

Long half-life in this class comes from structural modification that promotes albumin binding and resists enzymatic degradation. Weekly dosing is a consequence of the molecule, not a convenience decision.

Right.

This. The step interval is a floor, and reading it as a timetable is the most expensive mistake in the thread.

preclinical is not clinical and rodents are not small people

Adding the caveat the paper itself makes in its limitations section, which is stronger than anything in this thread.

GIP receptor biology is genuinely unsettled — there is a live argument about agonism versus antagonism at the receptor — and the clinical results are

This is the concept everything else on this board is downstream of.

Agreed on comparators. "Superior" means nothing until you know superior to what and at what dose.

comment on small win: dose escalation stopped being a problem at week 19 in c/retatrutide · 29 points · 1 year ago

Glucagon receptor agonism is associated with increased energy expenditure and with hepatic effects. That is a mechanistic story with strong preclinical support and limited phase 2 human data.

comment on tried weight for 3 weeks. here is what happened. in c/dosinglogs · 22 points · 1 year ago

Attribution requires that one thing change at a time. A log with dose, training and intake all moving in the same fortnight cannot assign anything to any of them.

Small fix: SURMOUNT is the obesity programme, SURPASS is the type-2 programme. They are different endpoints and the numbers do not transfer.

comment on [Discussion] the packaging advice in here is 2 years out of date in c/coldchain · 279 points · 1 year ago

Careful. "The cold chain failed" is a strong claim and the evidence you have is a warm box.

comment on [Question] how do you actually verify Mounjaro in c/tirzepatide · 456 points · 1 year ago

The four-week interval in the label is a minimum.

Not sure about this bit. The GIP arm being real does not tell you that it is what caused your particular week.

A token symbol identifies an asset, not a network. The same symbol commonly exists on several chains with incompatible address formats, and sending across that boundary is generally unrecoverable.

comment on [Discussion] gastric emptying is doing more work than we give it credit for in c/glp1science · 59 points · 1 year ago

Tolerance to the gastric effect develops with continued exposure while the appetite effect largely persists. That single fact explains most of the "it settles but it still works" pattern the side-effect board reports.

permalink

comment on what would you tell week-1 you about weekly update in c/dosinglogs · 146 points · 1 year ago

Correction: those two entries are 18 weeks apart, not consecutive, which changes how the middle of that series reads.

permalink

comment on help me understand handling, I have read the wiki twice in c/researchpeptides · -37 points · 1 year ago

Storage guidance that names a temperature, a state and a duration reflects actual stability work. Generic guidance usually reflects a template.

Choose a format you will still be able to open in two years. Plain text or a spreadsheet you export beats an application that may not exist.

Correction: that is glucose-dependent insulin secretion, which is why hypoglycaemia risk is low as monotherapy. Not the same claim as you made.

permalink

comment on three years of mechanism threads, summarised so you do not have to read them in c/glp1science · 102 points · 1 year ago

Glucose-dependent insulin secretion is why hypoglycaemia risk is low as monotherapy: the effect scales with glycaemia rather than acting unconditional

This is the concept everything else on this board is downstream of.

Research-use-only material is not approved for human use. Anything in this thread about how a research vial "should" titrate is a discussion about chemistry, not a plan.

comment on stalled for 7 weeks at 10mg and I refuse to panic this time in c/retatrutide · 1 points · 2 years ago

Phase 2 estimates effect and finds a dose range. Phase 3 estimates it precisely in a bigger population and catches what phase 2 was too small to see. Treating them as the same tier of evidence is the recurring error here.
