Contemporaneous records do not drift; reconstructed ones do, and the drift is systematic rather than random because memory smooths.

System suitability — repeat injections, tailing factor, plate count, RSD on area — is what tells you the instrument was fit for the measurement that day. Without it, the purity figure is unanchored.

What did week 13 look like compared with this one? The month either side is the useful comparison.

Went 10 to 12.5 on the calendar and had the worst fortnight of the whole run. Dropped back to 10 and everything settled.

SURMOUNT-1 ran 72 weeks with the top arm around 21% mean body weight change. SURPASS is the diabetes programme and reports glycaemic endpoints, so quoting the two interchangeably is a category error.

I would separate the two claims. That the GIP arm exists is uncontroversial; that it explains your food noise pattern is a guess.

That figure is the 68-week trial mean, not a weekly rate.

Adding one thing to this: the appetite effect and the scale run on separate clocks, so both halves can be true at once.

rasmus_petrescu is right — the integration choice is a decision and it should be stated alongside the result.

comment on 15mg — my 12-week log, condensed into one table in c/tirzepatide · 331 points · 1 year ago

The step schedule question, answered properly, because it comes up weekly.

The label sets a minimum interval of four weeks between increases. That is a floor on how fast you may go, and it exists because tolerability, not efficacy, is what limits most people. There is nothing in the pharmacology that says you must increase at four weeks, or at eight, or ever.

What decides it in practice is whether the effect you want is still there. If appetite is quiet and the trend is going the right way, the dose is doing its job. If both have genuinely gone flat for six weeks or more, that is a conversation worth having with someone who knows your history.

13kg over 42 weeks, never went past 10mg, and injection-site soreness stayed mild the whole way. Posting because the loud threads are all 15mg.

comment on [Vendor] QYB vs HJ on the same compound, same month — both cleared 99.0% in c/semaglutide · 17 points · 2 years ago

On the "everybody ends up at 2.4" claim, which comes up every few weeks.

The escalation schedule in the trials was designed to get people to a fixed study dose. Real use is not a trial. The dose that keeps your appetite quiet with side effects you can live with is the dose, and for a lot of people on this board that has been 1.7mg for a very long time.

The counter-argument, which is fair: some people genuinely do need the top of the range, and staying low out of caution costs them months. That is a conversation with someone who knows your history, not with us.

The system suitability argument, since it comes up whenever somebody posts a number without one.

Before the sample result means anything, the instrument has to be shown to be fit that day: replicate injections with an acceptable RSD on area, a tailing factor inside limits, adequate plate count, and a resolution check between the pair you care about.

None of that is exotic and all of it is routine in a laboratory that reports for a living. Its absence does not mean a number is wrong; it means the number is unanchored, and unanchored numbers should not be quoted to two decimal places on this board.

Disagree. 2.4 is the top of the label, not the goal, and "everyone ends up there" is just survivorship in the posts you read.
