comment on mg ÷ mL, that is the whole formula, why did i find this hard in c/reconstitution · 40 points · 2 days ago

Spent 10 weeks confused about this until somebody drew the table for me. 20mg, 2mL, 10mg/mL, 2.5mg is 25 units. That is the whole thing.

Which phase 2 arm are you quoting, and at what week?

comment on [Trial Data] TRIUMPH timeline — what we actually know vs what gets repeated in c/retatrutide · 6 points · 5 days ago

Right, and it bears repeating that nothing here is approved anywhere and research material is not for human use.

This is the sentence the rest of the board should read first. Phase 2 is not a label.

a U-100 syringe reads 100 units to the millilitre, every time

I would not round that figure. At 5mg/mL a rounding error of two units is a real fraction of the intended amount.

Adding more water does not change how much peptide is in the vial. It changes the concentration, which changes the volume you draw for the same amount. That is the entire relationship.

The phase 2 readout ran to 48 weeks in a few hundred participants with a slow escalation. Both the size and the duration matter when people quote the headline number.

phase 2 data only, and people quote it like it is a label

Small fix — a U-100 syringe is 100 units per mL. If yours is marked differently it is not U-100 and the whole calculation changes.

comment on help me understand mg/mL, I have read the wiki twice in c/reconstitution · 223 points · 1 months ago

Wrote the date on the vial as well as the concentration after losing track of which one was opened first.

comment on small win: glucagon stopped being a problem at week 32 in c/retatrutide · 126 points · 1 months ago

Stopped at a low step because there was no reason to go further and no data to tell me what further would do.

comment on how much of what we believe about bacteriostatic water actually comes from dead space threads in c/reconstitution · 49 points · 4 months ago

Cosigning the round-number approach. Reconstituting to something awkward is how you end up drawing 27.5 units at six in the morning.

permalink

comment on help me understand retatrutide, I have read the wiki twice in c/retatrutide · 11 points · 4 months ago

Cosigning the escalation point. Almost every difficult report on this board is from somebody who went up quickly.

permalink

comment on help me understand triple agonist, I have read the wiki twice in c/retatrutide · 62 points · 5 months ago

Same read. The energy-expenditure mechanism is the interesting bit and it is not established in humans at scale.

permalink

comment on why does nobody talk about concentration in c/reconstitution · 44 points · 5 months ago

Aim the stream down the wall of the vial rather than into the cake. Reconstitution is a solubility problem, not a mixing problem, and mechanical shear does nothing helpful.

Yes. Writing 10mg/mL on the vial takes four seconds and removes the entire class of error this board exists to fix.

Agreed on the heart-rate reports. They are consistent enough across the threads to be worth noting, not enough to be a finding.

How fast was the escalation? That is usually the variable that explains the reports.

permalink

The thing that surprised me was how much of the discussion here is inference rather than measurement.

permalink

comment on my A1c moved and I cannot work out whether TRIUMPH is why in c/retatrutide · 6 points · 9 months ago

Correction: TRIUMPH is the phase 3 programme. The number you are quoting is from the phase 2 readout, which is a different trial.

permalink

comment on [Question] how do you actually verify units in c/reconstitution · 323 points · 10 months ago

That only works if the fill weight is exactly the label weight, and it usually is not exactly anything.

permalink

comment on is it normal to get sulphur burps at week 87 in c/retatrutide · 13 points · 10 months ago

Glucagon receptor agonism is associated with increased energy expenditure and with hepatic effects. That is a mechanistic story with strong preclinical support and limited phase 2 human data.

Nothing containing this compound is approved anywhere. Research-use-only material is not approved for human use, and that is a statement of fact rather than a disclaimer.

permalink

comment on [Discussion] we are measuring TRIUMPH at the wrong time and calling it noise in c/retatrutide · 0 points · 11 months ago

Push back: quoting the phase 2 headline as an expected outcome is not a fair reading of a small trial with wide intervals.

Agreed — units are volume. Once that lands, the rest of the arithmetic is one division.

Increases in heart rate have been reported across this receptor class.

Adding the standing caveat: none of this is approved anywhere and research material is not for human use.
