Independent purity testing on this compound is thin compared with the older molecules, simply because fewer members have paid for it. Fewer results means wider uncertainty, not a verdict.

Sent a vial to VendorInvestigate out of curiosity. Result was 98.7% against a claimed 98.0%, which is the first independent number I had seen for this compound anywhere.

What is actually known, and what is being assumed.

Known: it is a small-molecule GLP-1 receptor agonist, dosed daily, with clinical programmes reporting in both obesity and type 2 diabetes. Its tolerability profile broadly resembles the class in the data published so far.

Assumed, frequently and confidently: that milligram comparisons with injectables mean something, that adherence is straightforwardly better because it is a tablet, that peptide purity discussions transfer to it. None of those hold. And nothing containing this compound is approved anywhere, which makes research-use-only material exactly that — not approved for human use.

This is the sentence the rest of the board should read first.

Agreed, and the glucagon arm is exactly why the comparison threads do not work.

Phase 2 estimates effect and finds a dose range. Phase 3 estimates it precisely in a bigger population and catches what phase 2 was too small to see. Treating them as the same tier of evidence is the recurring error here.

Two scans on the same machine 15 months apart. Lean mass held better than I expected and the lifts told the same story for free.

Because it is not a peptide, it is not subject to the same degradation pathways, does not require the absorption enhancers used for oral peptides, and can be formulated as a conventional tablet.

The thing that surprised me was how much of the discussion here is inference rather than measurement.

On means, which this board treats as targets and which are nothing of the sort.

A reported mean body weight change is the centre of a distribution that in these trials is very wide. Substantial numbers of participants did much better, and substantial numbers did considerably worse while remaining on the drug and in the analysis.

Quoting the mean as an expectation therefore misleads in both directions: it makes ordinary results look like failures and it makes exceptional results look normal. If a paper publishes the distribution — and several do, in the appendix — look at that instead. It is far more informative than the number in the abstract.

Cosigning on the glucagon arm. It is the mechanistic thread that connects the hepatic and the metabolic effects.

Titration is a tolerance ladder. Each step exists so the gastrointestinal effects settle before the next increase, which is why the intervals are what they are.
