same category: my cramps were sodium, not magnesium, and i spent £40 on the wrong mineral before checking anything.

Yes. The starting dose is a tolerance step. It is not supposed to do very much.

Correcting myself: that was a phase 2 readout and I described it as phase 3.

Went looking for independent testing on this and found essentially nothing, which was clarifying.
comment on [Discussion] non-peptide agonism is a genuinely different engineering problem in c/orforglipron · 25 points · 13 days ago
Oral semaglutide is a peptide co-formulated with an absorption enhancer and carries food and water timing requirements. A small molecule does not have that constraint, which is the practical distinction.

Appeal deadlines run from the date on the determination letter. They are strict, they are short, and a missed deadline usually forfeits that level entirely.

Analytically this is a small-molecule identity and purity problem, not a peptide one. The methods, the impurity classes and the reference standards are all different.

Agreed. The clone copies the front page and never the paperwork, because the paperwork is the expensive part.

Not convinced. That is a formulary exclusion rather than a prior authorisation denial, and the route to challenge it is different.
comment on help me understand week one, I have read the wiki twice in c/newbiequestions · 6 points · 4 months ago
The five things that would have saved me the most time in month one.
One: the first month is a tolerance check, not a results phase. Two: appetite usually changes before the scale does, so watch that instead. Three: write the concentration and the date on the vial the moment you reconstitute anything. Four: the loud posts are not the middle of the distribution. Five: nobody here — including whoever answers most confidently — is your clinician.
None of that is advice about what to take. It is the shape of the first few weeks, which is what I actually wanted and could not find.

comment on what changed for me between month 5 and month 9 in c/newbiequestions · -8 points · 6 months ago
How to ask a question here so that you get a useful answer.
Say what week you are in, whether you are using a pen or a vial, what country you are in, and what you have already read. Those four facts turn a question that gets three contradictory answers into one that gets a specific one.
And do ask. The basic questions get searched for constantly, so a clear answer here helps a stream of people who will never post. The only genuinely unwelcome thing on this board is being unpleasant to somebody for not knowing something yet.

Because it is not a peptide, it is not subject to the same degradation pathways, does not require the absorption enhancers used for oral peptides, and

The vocabulary, once, since half the questions here are really vocabulary questions.
Titration is stepping the dose up slowly for tolerance. Reconstitution is adding water to a freeze-dried powder. Bacteriostatic water contains a preservative; sterile water does not. COA is a certificate of analysis — a lab document about one batch. Area percent is a purity measure from that document, and it is not the amount of peptide in the vial. Units are a volume on an insulin syringe, not a dose.
That is most of the jargon on this site. The wiki has the longer version of each and it is linked in the sidebar.

Body weight varies by a couple of kilos day to day through fluid, glycogen and gut content, which is why a single weigh-in is noise and a fortnightly trend is signal.
comment on [Discussion] can we stop arguing about PDF metadata until somebody posts a number in c/scamalerts · 31 points · 10 months ago
The checks I run before sending money anywhere new, in order, and none of them cost anything.
Domain first, character by character, pasted rather than typed. Then registration age against the claimed trading history. Then a request, in writing, for a batch-specific certificate for the lot they would ship — not the catalogue document.
Then a small order, tested independently. Only after that a real one. This sequence has cost me a handful of small orders and it has never once cost me a large one, which is the whole point.
comment on [Discussion] can we stop arguing about PDF metadata until somebody posts a number in c/scamalerts · 55 points · 10 months ago
Domain age, registrar and certificate issuance date are all public and take about a minute to check. A site claiming years of history on a months-old domain is the cheapest signal available.

Titration is a tolerance ladder. Each step exists so the gastrointestinal effects settle before the next increase, which is why the intervals are what they are.
