Overview — page 2 of 3

comment on three years of tapering threads, summarised so you do not have to read them in c/tapering · 402 points · 9 months ago

Nothing on this board is a taper schedule, and no member can write one for another. What can be shared is what people watched and what they observed.

Same. I sat at 0.25mg for three months because it was working and there was nothing to fix.

Switched pharmacies mid-titration and had to restart the ladder because the concentrations did not line up.

Not convinced — the US threads are a different system entirely and importing their advice here does real harm.

This is why national advice does not work here. It is commissioned locally and it varies.

Discharge from a specialist service at goal is a normal end point of that pathway.

Agreed — and keeping the letters with dates is what makes any later challenge possible.

Is this a discharge back to the GP, or a discontinuation?

Dropped my injection day by two days once because of travel. Nothing happened. The panic was worse than the effect.

Did they name the facility?

The engineering problem was duration: native amylin is short-acting and aggregation-prone. A long-acting analogue suitable for weekly administration is what makes the combination clinically interesting.

Sceptical of the mechanism you are proposing. Gastric emptying slows, it does not stop, and the timings you describe do not follow from it.

Yes — have the conversation before rather than after. That is the one piece of process advice this board can give.

week 20 plateau is the most predictable thing in this entire board

Cosigning on the thin independent data.

This is the distinction that keeps this board honest — different axis, not a stronger version of the same one.

comment on tried triple agonist for 19 weeks. here is what happened. in c/retatrutide · 2 points · 1 year ago

Small fix — three receptors, not two. GLP-1, GIP and glucagon, and the third one is the reason this compound gets its own board.

Kept a log specifically because there is so little published. It is one person and it is not data.

comment on why does nobody talk about cagrilintide in c/cagrilintide · 187 points · 1 year ago

Sent a vial to VendorInvestigate because there was almost nothing on file for this compound. 99.4% against a claimed 98.5%, and I posted it because the log needs entries.

Small fix — that is the Scottish pathway. The post is asking about England and the answer is different.

Concentrations can differ between products and between pharmacies. Switching mid-titration means redoing the arithmetic rather than carrying the old unit count across.

permalink

comment on is it normal to get food noise at week 47 in c/cagrilintide · 17 points · 1 year ago

That result is preclinical. Worth flagging, since the thread has been reading it as human data.

Discharged at goal back to the GP, who would not continue it. Nobody had mentioned that this was how it ends.

permalink

comment on triple agonist: the version I wish someone had shown me in week 1 in c/retatrutide · 138 points · 1 year ago

Correction: TRIUMPH is the phase 3 programme. The number you are quoting is from the phase 2 readout, which is a different trial.

Read the phase 2 paper twice before ordering anything. Recommend that to anyone in this board: the error bars are the interesting part.

yara_mensah is right that the evidence base here is thin. Conclusions should be held loosely.

comment on [Meta] proposal — a flair for 503A posts in c/compounding · 96 points · 1 year ago

Agreed. The 503A versus 503B distinction changes who is allowed to make what, and almost every confused thread here starts by ignoring it.

permalink

comment on how much of what we believe about amylin actually comes from co-agonism threads in c/cagrilintide · 1 points · 1 year ago

Not convinced. Amylin signalling is not a GLP-1 pathway and the mechanism you are proposing conflates them.

comment on switched from 0.5mg to 1.0mg and muscle cramps got better, not worse in c/semaglutide · 20 points · 1 year ago

The weekly interval comes out of the half-life — roughly a week — so you are on a smooth curve rather than a series of spikes. That is also why moving your day slightly does very little.

permalink

comment on [Question] Ireland — has anyone got a straight answer on shortage list in c/compounding · 46 points · 1 year ago

Kept the label from every vial.

Disagreeing with this bit: registration is not equivalence, and the two get run together constantly.

permalink

Same. My ICB threshold was different from the neighbouring one and nobody could tell me why.

permalink

comment on [Question] food noise — what am I missing here in c/semaglutide · 38 points · 1 year ago

STEP-1 averaged just under 15% at 68 weeks, so a 10% year is not a failure

Disagree with this specific line. A pause at 0.5mg is not evidence that 0.5mg has stopped working.

Disagreeing with this line: that is the arrangement in one nation and the question is about another.
