comment on half-life is 168 hours. that is why your injection day barely matters. in c/glp1science · 1 points · 3 days ago

What was the exposure in that experiment relative to therapeutic?

area percent unless the report says otherwise

medutest_mel is right that the band matters more than the peak. Consistency across lots is the finding.

comment on why does everyone say week 4 is the wall in c/semaglutide · 7 points · 10 days ago

Small correction: 2.4mg is the maintenance ceiling on the label, not the starting maintenance dose. The distinction matters for anyone reading this later.

Correcting myself from earlier in the thread: I said week 9 and it was week 13. Same shape, wrong number.

Are you measuring anything other than weight? Waist is usually the one that keeps moving.

comment on oral 50mg vs injectable 2.4 — OASIS 1 numbers are closer than i expected in c/semaglutide · 78 points · 20 days ago

STEP-1 ran 68 weeks and landed just under 15% mean body weight change on 2.4mg. It is an average of a wide distribution, not a target you have missed.

comment on [Discussion] we need a standard template or these posts are unsearchable in c/labresults · 96 points · 1 months ago

The template is vendor, compound, batch, service, claim, result, and who paid. Every one of those fields exists because a result missing it has been argued about here before.

Adding the boring but useful bit: say whether you paid for it.

Disagreeing with this line: that gap is ordinary variance, not a contradiction between the lab and the certificate.

incretin effect first, then everything else in this board makes sense

comment on three years of half-life threads, summarised so you do not have to read them in c/glp1science · 155 points · 1 months ago

GIP receptor biology is genuinely unsettled — there is a live argument about agonism versus antagonism at the receptor — and the clinical results are robust regardless, which is an uncomfortable and interesting position.

Yes. The central component is the one that explains the reports on this site better than gastric emptying does.

permalink

comment on [Discussion] the telehealth advice in here is 3 years out of date in c/compounding · 24 points · 1 months ago

Nothing in this thread is medical advice, and the choice between arrangements is one for you and a prescriber who knows your history.

Disagreeing with this bit: registration is not equivalence, and the two get run together constantly.

comment on why does nobody talk about telehealth in c/compounding · 0 points · 1 months ago

the API source is the question nobody asks and everybody should

Adding one thing — ask for the facility name. Most will give it, and the ones that will not have answered you.

the label on a compounded vial is a legal document, read it

hydration_hank is right about the concentration trap. It breaks arithmetic that has been reliable for months.

sema is weekly for a reason, the half-life does the smoothing

Correction: that is regulatory approval, not coverage. They are separate decisions by separate bodies.

comment on [Discussion] we are measuring appetite at the wrong time and calling it noise in c/glp1science · 14 points · 2 months ago

Disagree.

Adding the caveat the paper itself makes in its limitations section, which is stronger than anything in this thread.

permalink

comment on [Question] how do you actually verify pharmacology in c/glp1science · 42 points · 3 months ago

Disagree.

the_poster_in_question_2026 is right that mechanism gives direction and not magnitude. Worth pinning.

The mental model, in four steps, that makes the rest of this site legible.

comment on [Question] how do you actually verify food noise in c/semaglutide · 139 points · 4 months ago

Steady state after a dose change takes four to five half-lives, so the fair judgement point on a new step is about a month, not four days.

Chain of custody is why member-funded testing is worth more than a supplied certificate: the vial tested is the vial that arrived at a member’s door.

Sceptical of the mechanism you are proposing. Gastric emptying slows, it does not stop, and the timings you describe do not follow from it.

permalink

Cosigning on GIP. The genuinely interesting thing is that the biology is not settled and the clinical result is nonetheless robust.

Yes. Approval and coverage are separate, and this board conflates them in about half its threads.

Glucose-dependent insulin secretion is why hypoglycaemia risk is low as monotherapy: the effect scales with glycaemia rather than acting unconditionally.
