mechanism — what am I missing here. I am not trying to be the "source?" guy. I would just like a source. Glucose-dependent insulin secretion is why hypoglycaemia risk…

comment on [Explainer] delayed gastric emptying does most of the work people credit to the brain in c/glp1science · 1 points · 2 days ago
Agreed — the incretin effect is the load-bearing concept and everything downstream reads differently once you have it.
comment on [Regain] 4kg back over a year and i am calling that a win in c/maintenancephase · 37 points · 15 days ago
Dose reduction and interval extension are different interventions. Halving the dose keeps the rhythm and lowers the level; stretching keeps the level and lowers the frequency, with a lower trough.
comment on [Long Term] what my labs look like three years in in c/maintenancephase · 12 points · 27 days ago
Wrote down what maintenance would look like at about week 20, before I needed it. Having decided in advance made the transition boring.
comment on [PSA] receptor is not what most of this community thinks it is in c/glp1science · 1 points · 2 months ago
mechanism explains a direction, not a magnitude
comment on dose stretching: the version I wish someone had shown me in week 1 in c/maintenancephase · 10 points · 2 months ago
Yes. The habits have to outlive the dose or the maintenance phase is just a countdown.
comment on dose stretching: the version I wish someone had shown me in week 1 in c/maintenancephase · 42 points · 2 months ago
Push back: that is not a plateau, that is maintenance. You have arrived and you are describing it as a failure.
comment on incretin: what the trials say vs what this community says in c/glp1science · 33 points · 2 months ago
receptor agonism is not the same as receptor activation in every tissue
comment on 12 months in and orforglipron is still the thing I get wrong in c/orforglipron · 11 points · 2 months ago
daily dosing changes adherence in both directions
comment on am I the only one who found long term harder than the injections in c/maintenancephase · 10 points · 3 months ago
the habits are the part that has to outlive the dose
comment on ACHIEVE: what the trials say vs what this community says in c/orforglipron · 31 points · 3 months ago
Right, and comparing milligrams between a small molecule and a peptide is meaningless in either direction.
comment on [Meta] the mechanism rule is doing its job and people should stop complaining in c/glp1science · 30 points · 4 months ago
Agreed that receptor distribution is the key to the side-effect map. It is not a mystery, it is anatomy.
comment on what would you tell week-1 you about maintenance in c/maintenancephase · 1 points · 4 months ago
plan the maintenance phase before you need it
comment on appetite: what the trials say vs what this community says in c/glp1science · 274 points · 5 months ago
GIP is the arm people argue about because the biology is genuinely unsettled
comment on [Lab] 15th independent test on WXT — 99.4% on a claimed 98.5%, and the trend is the interesting part in c/orforglipron · 13 points · 8 months ago
Daily dosing gives a very different exposure profile from a weekly injectable: peaks and troughs within each day rather than a smoothed weekly curve.
comment on [PSA] gastric emptying is not what most of this community thinks it is in c/glp1science · 10 points · 9 months ago
Correction: that is glucose-dependent insulin secretion, which is why hypoglycaemia risk is low as monotherapy. Not the same claim as you made.

What does the discussion section say about the limitation you are glossing?
comment on [Question] mechanism — what am I missing here in c/glp1science · 5 points · 9 months ago
Asked a question here that I thought was stupid and got three papers back. Best thread I have been in on this site.
comment on oral GLP-1 — 12 things I got wrong before I got it right in c/orforglipron · 0 points · 10 months ago
Daily dosing, so what does the exposure profile look like across the day?
Adding the standing caveat — unapproved, and research material is not for human use.
comment on [Question] how do you actually verify maintenance in c/maintenancephase · 59 points · 11 months ago
Withdrawal studies in this class consistently show substantial regain after discontinuation. That is a pharmacological finding about a chronic condition, not a statement about anyone’s discipline.
comment on why does nobody talk about incretin in c/glp1science · 152 points · 1 year ago
Does the effect persist with continued dosing or does tolerance develop?
Agreed — and it is why the central and peripheral stories are complementary rather than rival.
comment on stalled for 3 weeks at 0.5mg and I refuse to panic this time in c/maintenancephase · 411 points · 2 years ago
regain after stopping is the expected outcome, not a personal failure
comment on how much of what we believe about regain actually comes from lowest effective dose threads in c/maintenancephase · 250 points · 2 years ago
Not convinced. Interval stretching changes your exposure profile, not just the total, and the two are not equivalent.
comment on does receptor actually matter or is it forum lore at this point in c/glp1science · 16 points · 2 years ago
What was the exposure in that experiment relative to therapeutic?
comment on [Question] small molecule — what am I missing here in c/orforglipron · 1 points · 2 years ago
That is a phase 2 result being quoted as though the programme had reported.
Adding the standing caveat — unapproved, and research material is not for human use.

Which programme and which readout are you quoting?
Adding the standing caveat — unapproved, and research material is not for human use.

Yes. The habits have to outlive the dose or the maintenance phase is just a countdown.
