comment on what would you tell week-1 you about non-peptide in c/orforglipron · 10 points · 12 months ago
Disagree. You are comparing doses across a small molecule and a peptide, which is not a comparison of anything.
comment on the half-life question that gets asked weekly, answered properly in c/glp1science · 1 points · 12 months ago
Read the primary paper after arguing about the summary for a fortnight. The discussion section said almost the opposite of what the thread had concluded.
comment on am I the only one who found tracking harder than the injections in c/dosinglogs · 4 points · 1 year ago
Added a sleep column late and immediately understood two of my worst weeks.

nobody owes anybody a starting weight
comment on [Results] 51 weeks on 2.4mg, full numbers, ask me anything boring in c/orforglipron · 139 points · 1 year ago
This. The absence of food and water timing restrictions is the practical difference people will actually notice.
comment on [Discussion] the appetite advice in here is 3 years out of date in c/glp1science · 61 points · 1 year ago
GLP-1 receptor agonism acts both peripherally — insulin secretion in a glucose-dependent way, slowed gastric emptying — and centrally, on appetite regulation. The central component is the better explanation for sustained intake reduction.

a log with no dates is a story

Disagree with daily weighing. You are logging fluid, and the emotional cost is real for very little information.
comment on what would you tell week-1 you about ALT in c/bloodwork · 16 points · 1 year ago
Disagree with reading anything into that. One value, one morning, and you changed two other things in the same window.
comment on [Discussion] progress is doing more work than we give it credit for in c/progresspics · 310 points · 1 year ago
Why this board is text only, since new members ask every week.
A photograph invites comparison on things nobody can control — lighting, posture, where you carry weight, what you looked like before any of this. Text invites description of what changed for you: the jacket, the stairs, the measurement, the month it took.
The result is a board where the posts are about people’s actual experience rather than about images, and where nobody is competing on the wrong axis. It was a deliberate choice, it has been challenged periodically, and every time it has been kept.
comment on [Lab] QYB orfo — Medutest came back 99.2% against a claimed 99.0% in c/orforglipron · 290 points · 1 year ago
Is that from the publication or the press release?
comment on baseline labs: the version I wish someone had shown me in week 1 in c/bloodwork · 89 points · 1 year ago
Substantial weight loss independently moves lipids, liver enzymes and several other markers. Attributing a change to a compound while losing weight is confounded by design.
comment on [Discussion] dose history is doing more work than we give it credit for in c/dosinglogs · 3 points · 1 year ago
Right, and the payoff arrives around month six when a pattern becomes visible that nobody could hold in their head.
comment on my TSH moved and I cannot work out whether ACHIEVE is why in c/orforglipron · 4 points · 1 year ago
What did the tolerability table look like at that dose?
comment on my TSH moved and I cannot work out whether ACHIEVE is why in c/orforglipron · 19 points · 1 year ago
Correction: that is the oral peptide product, which is a different thing entirely. This one is a small molecule.

the tolerability profile reads broadly similar to the class
comment on reading ALT threads from 2024 and half of it aged badly in c/bloodwork · 146 points · 1 year ago
Repeat testing before acting is standard practice for a reason: regression to the mean does a lot of work on single outlying values.
comment on reading oral GLP-1 threads from 2024 and half of it aged badly in c/orforglipron · 3 points · 1 year ago
It is a non-peptide small-molecule agonist at the GLP-1 receptor. That is a substantial medicinal chemistry achievement: getting a small molecule to activate a receptor evolved for a peptide ligand.
comment on milestone — 2 things I got wrong before I got it right in c/progresspics · 27 points · 2 years ago
Text-only posting removes the comparison mechanism that makes image boards difficult, which is why this community adopted the rule and has kept it.
comment on does receptor actually matter or is it forum lore at this point in c/glp1science · -36 points · 2 years ago
Why mechanism talk keeps misleading people, including me.
A mechanism tells you a direction. It does not tell you a magnitude, a timescale, or whether the pathway is operative at the exposures involved. "Receptor X is expressed in tissue Y" is a fact; "therefore effect Z in a person" is a hypothesis with several missing steps.
The corrective is boring and it works: ask whether the evidence is preclinical or human, ask what exposure was used, and read the limitations section before the abstract. Most of the confidently wrong posts on this board — several of them mine — skipped all three.
comment on [Question] small molecule — what am I missing here in c/orforglipron · 13 points · 2 years ago
Spent an evening reading about small-molecule agonism at a peptide receptor. Genuinely interesting engineering.

Does the effect persist with continued dosing or does tolerance develop?
comment on [Discussion] incretin is doing more work than we give it credit for in c/glp1science · 38 points · 2 years ago
That study was in a rodent model. Worth stating, since the thread has been reading it as human data.

Correction: that is glucose-dependent insulin secretion, which is why hypoglycaemia risk is low as monotherapy. Not the same claim as you made.
