Area percent is the integrated area of your peak over the total integrated area at one wavelength on one gradient. Change any of those and the number changes without the sample changing.

Not convinced by that integration. Dropping the baseline there absorbs part of the shoulder into the main peak.

comment on someone explain HPLC to me like I have not read a paper in years in c/hplc · 549 points · 1 year ago

Sent the same vial to Medutest and PeptideMeter. 98.4% against a claimed 98.0%. The difference was the gradient, not the material.

baseline choice is a decision, not a measurement

This.

Agreed. And the corollary is that the method line on a certificate is not decoration, it is the number’s provenance.

How old is the column and roughly how many injections has it seen?

a blank injection between samples costs four minutes and settles most arguments

comment on unpopular opinion: most of what gets said here about compounding is guesswork in c/glp1aus · 59 points · 1 year ago

The compounded arrangements changed while I was mid-thread and I was arguing from a superseded position.

Disagree. Severe abdominal pain is not on the list of things to wait out, and this board should not be treating it as one.

reproducibility beats resolution if you only get one of them

That reads like advice, and none of us can give it. Describe what you did, not what somebody else should do.

comment on help me understand heart rate, I have read the wiki twice in c/sideeffects · -14 points · 1 year ago

Most of the common complaints trace back to slowed gastric emptying — early fullness, reflux, nausea after a large or fatty meal. That is also why meal size and speed change the experience more than anything else.

Method questions stay here; supplier claims go to c/vendorvetting with a document attached.

mass_spec_maggie is right — the integration choice is a decision and it should be stated alongside the result.

Gave the same trace to two people I trust and got two integrations about a point apart. That was the most educational afternoon I have had here.

Disagreeing with this one line: that symptom is not on the wait-and-see list and should not be posted as though it is.

ipamorelin_i is right about the timing. The day-of-week pattern is the diagnostic bit, not the severity.

comment on [Question] special authorization — what am I missing here in c/glp1canada · 138 points · 1 year ago

A rejected claim at the pharmacy counter and a denied authorization are different events with different remedies. The first can be an administrative or quantity issue; the second is a determination you can appeal.

Push back: a longer run is not automatically better resolution. You are trading peak width for time and the ratio is what matters.

comment on three years of special authorization threads, summarised so you do not have to read them in c/glp1canada · 0 points · 2 years ago

Employer-sponsored plans have their own criteria and appeal processes, independent of the provincial formulary. Which one applies determines everything about the route.
