dead space in the needle hub is real and it is small

What concentration did you actually reconstitute at, rather than what did you intend to?

Cosigning the beyond-use date question. What it is based on tells you whether anybody has done stability work.

Not convinced by the linearity assumption. Dose-response in this class is not linear and the trials show it.

Push back: PCOS as a diagnosis and insulin resistance as a finding are not interchangeable, and the thread is using them as one.

Asked for the beyond-use date basis and got a real answer with a stability reference attached. Not universal, apparently.

Concentration on the compounded vial was different from what I had been using and I nearly did the arithmetic on autopilot.

Pushback on the syringe advice: a U-100 barrel reads units of volume. Calling them "doses" is how the confusion starts.

tolerance to the gastric effect develops, appetite effect largely persists

glucagon agonism sounds paradoxical until you read the energy expenditure work

comment on why does nobody talk about units in c/reconstitution · 13 points · 2 years ago

I would not round that figure. At 10mg/mL a rounding error of two units is a real fraction of the intended amount.

comment on [Question] how do you actually verify appetite in c/glp1science · 94 points · 2 years ago

Tolerance to the gastric effect develops with continued exposure while the appetite effect largely persists.

Agreed — and it is why the central and peripheral stories are complementary rather than rival.

I would not round that figure. At 5mg/mL a rounding error of two units is a real fraction of the intended amount.

comment on [Discussion] incretin is doing more work than we give it credit for in c/glp1science · 109 points · 2 years ago

Agreed — the incretin effect is the load-bearing concept and everything downstream reads differently once you have it.

comment on unpopular opinion: most of what gets said here about concentration is guesswork in c/reconstitution · 0 points · 2 years ago

The arithmetic in full: concentration is mass over volume, so 20mg into 1mL gives 20mg/mL.

aurel_boateng has the units point exactly right. They are a volume on the barrel and nothing else.
